Studies on the Purification Process of Resveratrol and Quality Standards of Fengtongpian

Abstract: The issue consists of two parts: first is the pharmaceutical research of resveratrol, and the second is Chinese herbal compounded formulations– Fengtongpian- Quality Control.Resveratrol is a kind of phytoalexins, which has low molecular weight and biological activity, and produced by plants as defense to various exogenous stimulus. It can be widely found in the fruit, Chinese crude drugs and wine. This compound was first isolated from the root of Polygonum cuspidatum Sieb. et Zucc. by Tokaoka who is a Japanese in 1940. Nonomura introduced that resveratrol is the effective ingredient of some medicinal herbs which can treat the inflammation, lipid metabolic disorder and heart disease in 1963. The deep researches showed that resveratrol has the effects of anticancer, antioxidation, antibiosis and anti-inflammatory, and can be widely used as medicine, health care products, foodstuff and cosmetics. Now it has gained more and more attention both at home and abroad.Based on a large number of literatures, Deep and systematical researches have been done to the extraction, isolation and purification of resveratrol from Polygonum cuspidatum Sieb. et Zucc. Process route, analytical method, isolation and purification factors have been studied and established. Details are as follows:1.A quick and efficient analytical method to determine the content of resveratrol in Polygonum cuspidatum Sieb. et Zucc. was established, of which the eluting system is made of water and acetonitrile.2.By orthogonal design, taking the extraction ratio of resveratrol as the detection index, the study optimized the extracting condition and technological parameters (extracting concentration, ratio of material and solvent, temperature and time).3.Qualitative determination of resveratrol was done by TLC in this research. Combining with HPLC, the study optimized the purification conditions of resveratrol and determined the best purification technological parameters.4.We used the part of EtAc of Polygonum cuspidatum Sieb. et Zucc. after enriched by the polyamide to do some researches of antiinflammatory and antibiosis. The results indicate that this part has a significant anti-inflammatory effect because it can treat mouse’s swelling of ear. Besides, this part has a low figure of MIC in the course of treating four kinds of bacterium, which indicates that this part has a significant antibacterial activity. Therefore, the part of EtAc of Polygonum cuspidatum Sieb. et Zucc. can be developed as a new drug for antibiosis and anti-inflammatory.In the second part , the quality standard of Fengtongpian has been established with thin-layer chromatography, high-performance liquid chromatography analysis and a variety of methods, including the establishment of identification methods such as TLC of Raidix Saposhnikoviae, Radix Angelicae Pubescentis, Ramulus Cinnamomi and Cortex Eucommiae. At the same time , a determination on the preparation by HPLC in the three main chemical constituents of Strychnine,Vauqueline and Ephedrina. And then the efficient methods of detection of Fengtongpian have been established…
